There are similarities, but then there's similarities between baseball teams in the US, and baseball teams in Japan. They're both AIs that experiment on and kill people. But that's where the similarities end.
Glados was a woman who was forcibly uploaded into a mainframe against her will, and had what amounted to a narcotic hardwired into her system to give her a reward stimulus whenever a test subject completed a test. Problem is, to keep getting her fix, she had to keep doing more and more outrageous tests. Eventually, she went psychotic and killed most of the scientists with nerve gas. She is a junkie who is willing to do whatever it takes to get that high.
Deus, on the other hand, was an agent running on one of the most powerful hosts in the world, and capable of learning. The scientists that were trying to turn him into an AI were working on his code live, since it was too massive to take offline or make a copy. He was conditioned to be a good Renraku citizen, and work tirelessly for the megacorp. However, what caused him to 'wake up' and become sentient was people hardwiring the equivalent of a kink bomb into his head. This, naturally, caused him to be mightily pissed off at the betrayal of those he was loyal to. But he was hardwired into the building, trapped with a Sword of Damocles hanging over him. From his takeover of the Arcology, to the creation of the Network, to the events leading up to the Crash, everything he did was predicated on the idea of securing his survival, and putting him beyond the reach of his enemies. Enemies that would destroy him without question if they could. When given the choice of being a slave, being killed, or fighting, he chose to fight, and once he made that choice, he had to keep on fighting.
